dc.description.abstract | La dissociazione della proteina tetramerica transtiretina (TTR, trasportatore plasmatico della tiroxina) in monomeri e l’aggregazione patologica dei monomeri in fibrille rappresentano passaggi cruciali nelle amiloidosi da TTR. Piccoli composti organici di sintesi interagiscono specificamente con i siti di legame della tiroxina della TTR, stabilizzandone lo stato tetramerico nativo e conseguentemente inibendo la fibrillogenesi in vitro. In questo studio abbiamo verificato che anche alcuni polifenoli naturali e loro metaboliti sono in grado di legarsi alla TTR, stabilizzandone lo stato nativo ed inibendo in vitro la fibrillogenesi. Questi composti sono quindi di potenziale interesse per la terapia farmacologica delle amiloidosi da TTR. | it |
dc.description.abstract | The dissociation of tetrameric human transthyretin (TTR) into monomers and monomer misfolding and misassembly are believed to represent crucial steps leading to pathologic formation and extracellular deposition of amyloid fibrils. By specifically interacting with the TTR thyroxine-binding sites, small synthetic organic molecules are able to stabilize the tetrameric native state of TTR, inhibiting in vitro fibrillogenesis. We show here that some natural polyphenols, and some of their metabolites, are also able to bind with high affinity to TTR, protecting its native state and inhibiting in vitro fibrillogenesis. Therefore, such compounds represent promising drugs for the therapy of TTR amyloidoses. | it |
