Synthesis of hybrid systems for biomedical applications by the functionalization of nanostructures

Abstract

In questo lavoro, è stata studiata la sintesi di nanosistemi ibridi solubili in acqua capaci di ottenere un effetto potenziato di terapia fotodinamica a seguito di un irraggiamento con raggi X. Per raggiungere questo scopo, nanoparticelle d'oro (AuNPs) sono state coniugate a un fotosensibilizzante (porfirina) per ottenere un aumento di fosforescenza (MEP) e una maggiore generazione di 1O2. La (4-idrossifenil)porfirina (THPP) è stata monofunzionalizzata con leganti tiolo terminali di diversa lunghezza e coniugata sulla superficie delle AuNPs. Infine, la produzione di 1O2 è stata valutata mediante l'utilizzo di ADPA a seguito ad un irraggiamento con luce bianca. Rispetto alla THPP libera, il coniugato NS3 ha prodotto un aumento di 1O2 sia con luce bianca che con raggi X. Al fine di migliorare la stabilità in vivo e l’uptake cellulare del nanosistema, è stata studiata preliminarmente l'incorporazione di NS2 nei liposomi. La loro sintesi è stata eseguita presso il Technion (Israele) mediante il metodo “thin film layer” ottenendo una buona efficienza di incapsulamento. Durante questo periodo all'estero, è stato anche studiato un nuovo delivery system capace di raggiungere selettivamente le cellule cancerogene del polmone per indurre il silenziamento della mutazione del gene KRAS attraverso la tecnologia chiamata “RNA interference” (RNAi). ​

In this work, the synthesis of water soluble hybrid nanosystems was investigated to achieve an X-Ray enhanced Photodynamic therapy (PDT) effect. According with this, Gold Nanoparticles (AuNPs) were conjugated to a photosensitizer (porphyrin) to obtain metal-enhanced phosphorescence (MEP) and an increased 1O2 generation (SOG). (4-hydroxyphenyl)porphyrin (THPP) was monofunctionalized with thiol ending linkers with different length and conjugated on AuNPs’ surface. Finally, the 1O2 production was evaluated through the use of ADPA under a white light irradiation. Compared to the free THPP, the conjugate NS4 produced an enhancement of 1O2 both under white light and X-Rays.In order to improve the in vivo stability and cellular uptake of the nanosystem, the preliminar incorporation of NS1 into liposomes was investigated. Synthesis of NS2 liposomes was performed at the Technion (Israel) through the thin film layer method achieving a good encapsulation efficiency. During this abroad period, was also investigated a novel delivery system that selectively goes in lung cancer’s cells to induce the silencing of KRAS mutation through the RNA interference (RNAi) technology. ​
