Abstract: La fertilità è un carattere di fondamentale importanza per l’economia aziendale dell’allevamento bovino da latte. Si hanno risvolti negativi quando performance riproduttive sono inefficienti o non rispondenti alle esigenze attese, a fronte degli ingenti sforzi economici messi in atto, i quali difficilmente potranno essere ammortizzati con una gestione non ottimale della riproduzione. L’industria zootecnica usa sempre più frequentemente tecniche di riproduzione assistita per la produzione di embrioni in vivo tramite Multiple Ovulation Embryo Transfer (MOET) per effettuare Embryo Transfer (ET) o produzioni embrionali in vitro tramite di In Vitro Fertilization (IVF) per l’attività di Ovum Pick Up (OPU). Lo studio tramite approcci genome-wide association studies (GWAS) dei caratteri rilevati tramite tecnica ET e OPU è stata utilizzata in questo elaborato di tesi per determinare quali regioni del genoma bovino potessero essere associate a questi caratteri di fertilità. Tramite lo sviluppo di un modello statistico è stato inoltre possibile valutare quali aspetti ambientali e dell’animale avessero un effetto statisticamente significativo durante la raccolta delle osservazioni in campo e quindi durante l’attività di produzione e trapianto embrionale. Lo studio, durato 3 anni, ha messo in mostra come le pluripare determinano una produzione embrionale quantitativamente maggiore delle manze, ma qualitativamente inferiore a quest’ultime, mentre la qualità oocitaria per l’aspirazione follicolare potrebbe essere influenzata dallo status in cui si effettua l’operazione. Inoltre, i risultati preliminari dello studio GWAS hanno identificato una regione nel cromosoma 11 come potenzialmente associata alla maturazione embrionale; probabilmente per una regione sul cromosoma 10 si potrebbe avanzare un ipotetico ruolo regolatore sulla qualità del materiale embrionale, mentre due regioni sul cromosoma 1 e 4 sembrerebbero svolgere attività di maturazione follicolare. Ulteriori approfondimenti sono necessari per valutare il potenziale utilizzo della selezione genomica su questi nuovi caratteri di fertilità identificati grazie a queste tecniche di riproduzione assistita.
Fertility is a trait of fundamental importance for the business economy of dairy cattle breeding. There are negative implications when reproductive performance is inefficient or does not meet expectations, in the face of the enormous economic efforts put in place, which can hardly be amortized with a non-optimal management of reproduction. The livestock industry increasingly uses assisted reproduction techniques to produce embryos in Vivo through Multiple Ovulation Embryo Transfer (MOET) to carry out Embryo Transfer (ET) or embryonic productions in Vitro through In Vitro Fertilization (IVF) for the Ovum Pick Up (OPU) activity. The application of genome-wide association studies (GWAS) on those new phenotypes collected with ET and OPU techniques was considered to determine which region of the bovine genome could be associated to fertility. Via the development of a statistical model, it was also possible to evaluate which animal and environmental factors influenced the traits during the collection of observations in the field and during the activity of production and embryonic transplantation. The study, which lasted 3 years, has shown how the multiparous cows produce a quantitatively greater embryonic production than the heifers, but qualitatively lower than the latter, while the oocyte quality for follicular aspiration could be influenced by the status in which the operation was performed. Furthermore, the results could indicate that a region on BTA11 is potentially responsible for embryonic maturation, and probably for a region on BTA10 a hypothetical regulatory role on the quality of the embryonic material could be advanced, while two regions on BTA1 and BTA4 seem to carry out follicular maturation activity. Further studies are needed to evaluate the potential application of genomic selection for those novel phenotypes associated to fertility
