Protein-based systems for production and detection of reactive oxygen and nitrogen species

Abstract

Questo lavoro si è concentrato sullo studio di alcune proteine globulari al fine di mostrarne le versatili potenzialità come strutture di dimensioni nanometriche in grado sia di trasportare specie reattive sia di rivelare molecole attive a livello cellulare. Per quanto riguarda lo studio di proteine come trasportatori di specie reattive, l’attenzione si è focalizzata sulla caratterizzazione di agenti teranostici, ovvero di nanocarrier in grado di trasportare sia molecole per la localizzazione non-invasiva della malattia e del farmaco, sia agenti per la cura al livello cellulare. Nello specifico, si sono individuati carrier proteici in grado di legare e veicolare un fotosensibilizzatore (PS), ovvero una molecola che in seguito all’assorbimento di fotoni ha un’elevata probabilità di passare in uno stato di tripletto da cui, per energy transfer, può produrre ossigeno singoletto, una specie reattiva non radicalica in grado di indurre la morte di cellule tumorali o batteriche. Uno dei requisiti fondamentali affinché un PS possa essere utilizzato con successo nella terapia fotodinamica antitumorale (PDT) o antimicrobica (aPDT) è che sia solubile e che si possa localizzare nelle specifiche zone di interesse. Nell’ottica di sviluppare sistemi biocompatibili si sono utilizzati PS di origine naturale, come Ipericina o Curcumina estratti rispettivamente dalle piante di Hypericum perforatum e di Curcuma longa. Un altro sistema fotosensibilizzante studiato è stato la zinco mioglobina (ZnMb), una mioglobina il cui gruppo eme è stato sostituito con Zinco protoporfirina IX (ZnPP-IX), un noto fotosensibilizzatore usato in PDT. Affinché un PS possa essere utilizzato in terapia fotodinamica deve essere solubile in soluzione acquosa. Tuttavia, l’aggregazione per idrofobicità è uno dei problemi comuni per diversi PS. Da qui la necessità di utilizzare complessi proteici per trasportare i PS, evitare la loro aggregazione e veicolarli nelle immediate vicinanze del target. Seguendo questo approccio, sono state studiate diverse proteine globulari capaci di legare PS di origine naturale. La seconda parte di questo studio riguarda la possibilità di usare proteine come biosensori per molecole, ioni o secondi messaggeri. In particolare l’attenzione è stata concentrata sullo sviluppo di sistemi fluorescenti in grado di rivelare la presenza di ossido nitrico (NO•) sia in vitro che in vivo. NO• è un gas radicalico altamente reattivo prodotto da varie cellule negli organismi viventi. La sua sintesi è mediata dalla Nitrossido Sintasi (NOS), un complesso enzimatico presente in 3 forme: neuronale (nNOS), endoteliale (eNOS) e inducibile (iNOS). Le prime 2 sono costitutive e producono NO• in basse quantità (viene generato ossido nitrico in concentrazioni nell’ordine del nanomolare) affinché agisca da mediatore fisiologico; la forma inducibile viene espressa solo in casi particolari, ad esempio nei momenti in cui viene stimolato il sistema immunitario, e produce NO• in abbondanza (si raggiunge una concentrazione pari a decine di micromolare a seconda dello stimolo), per far sì che possa contribuire al ripristino dell’omeostasi ed all’eliminazione del patogeno che sta minacciando la salute dell’organismo. NO• ha un’azione paracrina e svolge una svariata serie di effetti, a seconda dell’isoforma enzimatica che lo produce e della cellula su cui agisce. A livello del sistema cardiovascolare l’attività principale è quella vasodilatatrice ed antiaggregante. Nel sistema nervoso modula la trasmissione nervosa a livello del SNC e nei plessi nervosi periferici non adrenergici e non colinergici, soprattutto del tratto gastrointestinale, e dei corpi cavernosi. Il suo compito nel sistema immunitario è quello di difesa dell’ospite tramite l’effetto citotossico. Nonostante siano state proposte diverse metodologie per la quantificazione dell’NO•, il monitoraggio intracellulare in tempo reale della produzione di NO• con metodi non invasivi è un problema ancora aperto. A questo riguardo, lo sviluppo di sensori fluorescenti codificati geneticamente appare una strategia particolarmente promettente. L’idea di questo progetto è stata quella di identificare proteine fluorescenti le cui proprietà di emissione (intensità e tempi di vita di fluorescenza) risentano dalla concentrazione di NO• presente in soluzione. L’attenzione si è concentrata su proteine mutanti della Green Fluorescence Protein (GFP), come mTag BFP2, TagRFP, EYFP, EGFP, mCherryFP and CiNP-mTagBFP2. Quest’ultima si tratta di un costrutto chimerico creato dalla fusione di una proteina fluorescente nel blu e una Nitroforina di Cimex. La Nitroforina, grazie alla presenza del gruppo eme, è in grado di legare NO•, quando il ferro si trova nello stato Fe3+, e questa interazione determina una variazione nello spettro di assorbimento. Si è pensato, quindi, di sfruttare questo cambiamento spettrale per favorire un meccanismo di Förster Resonance Energy Transfer, FRET, all’interno di questo costrutto chimerico. Il fenomeno di trasferimento non radiativo di energia è stato rivelato osservando una diminuzione del tempo di vita della fluorescenza del donatore, ovvero della proteina fluorescente. In questa ricerca si è sottolineata la versatile potenzialità di proteine globulari utilizzabili sia come trasportatori di PS per applicazioni in PDT e aPDT sia come biosensori a livello intracellulare in microscopia di fluorescenza.

This work focused on the study of globular proteins in order to define their versatile applications both as nanostructures able to delivery reactive species and to monitor active molecules at cellular level. With regard to the investigation of transport proteins, the attention was focused on compounds that can self-assemble thanks to their physical-chemical properties. In these cases, the hydrophobicity of the active site of a protein can be exploited to assemble nanostructured compounds that have relevant biotechnological uses. This approach has applications in drug delivery, in which a protein based carrier can transport photo-physically active molecules. These complexes can be used as carriers of therapeutic or diagnostic active compounds and, if properly engineered, they can have both functionalities and assume theranostic characteristics. In particular, one of the purposes of this work is to study and verify if these complexes can have future applications in photo-treatments to kill cancer cells, Photodynamic Therapy (PDT), or eradicate microbial infections, Antimicrobial Photodynamic Therapy, (aPDT). The idea behind this project was to identify and characterize proteins able to carry a photosensitizer (PS). A PS is a molecule, which upon absorption of a photon, is able to switch with high probability from the ground state to a triplet state. In the triplet state a PS can interact with molecular oxygen and generate singlet oxygen, a reactive and cytotoxic oxygen species. In view of developing biocompatible systems, our attention was focused on naturally occurring PSs. In this respect, PSs extracted from the plant Hypericum perforatum and from Curcuma longa, respectively, Hypericin (Hyp) and Curcumin (Cur), were analysed. In addition to these molecules, another naturally occurring photoactive system, Zinc Protoporphirin IX (ZnPP-IX), was examined. This last photosensitizing system, in contrast to the previous, is characterised by a covalent bond between PS and protein matrix, the apomyoglobin. One of the fundamental prerequisites for applying a PS in PDT or in aPDT is the solubility of molecules. The hydrophobic aggregation is a common problem for different PSs, therefore, to avoid this situation, it is possible to use protein structures to bind, solubilize and transport the PS to specific cells. Pursuing this approach, globular proteins capable of carrying naturally occurring photoactive molecules for PDT or aPDT applications were studied. The other study performed in this work concerns the possibility to use proteins as biosensors of molecules, ions or second messengers. In particular, the attention was focused on the development of biological fluorescence systems able to monitor nitric oxide (NO•) concentration both in vitro and in vivo. NO• is a free radical, involved in different processes in bacteria, plants and mammals. In particular, thanks to its high reactivity, NO• plays an important role in nervous, immune and cardiovascular system. It is therefore evident that is a necessary to develop methods able to detect this messenger molecule, both in vitro and in vivo. Owing to the inherent importance of monitoring NO•, several methodologies have been developed, but in general, they suffer from the lack of capability for intracellular detection. The purpose behind this project is to identify fluorescent proteins that change their emission properties (intensity and/or lifetime) depending on NO• concentration in order to use them as NO•-sensor in intracellular fluorescence microscopy. This new approach has been taken into consideration in this thesis and, in particular, an unexpected sensitivity for different fluorescent proteins has been studied and found. The attention was focused on Green Fluorescent Protein (GFP) mutated proteins, like mTagBFP2, TagRFP, EYFP, EGFP, mCherryFP and CiNP-mTagBFP2, a chimeric protein created by the fusion of Cimex Nitrophorin protein and blue fluorescent protein. In the latter protein, the presence of NO• causes a variation in the absorption spectrum that facilitates a Förster Resonance Energy Transfer (FRET) between the heme and the fluorescent proteins. It was possible to monitor the presence of this mechanism observing the reduction in the fluorescence intensity and lifetime of the fluorescent protein. In this research is evident a versatile potentiality of globular proteins usable both as carrier of PSs for applications in PDT and aPDT and as biosensors for the intracellular detection of NO•.
