Evaluation of nutritional quality of cereals: bioaccessibility and effect of thermal treatment

Abstract

L’evidenza epidemiologica suggerisce che il consumo di cereali integrali sia associato ad un ridotto rischio di malattie cronico-degenerative, probabilmente a causa del loro contenuto in fibra alimentare, micronutrienti e composti antiossidanti, localizzati principalmente negli strati esterni della cariosside. La qualità nutrizionale di un alimento tuttavia non dipende soltanto dal contenuto in composti bioattivi, ma anche dalla loro bioaccessibilità, che è influenzata dalla struttura della matrice e dai trattamenti (termici, meccanici) che l’alimento subisce. Da queste considerazioni, il progetto di dottorato si è sviluppato in due tematiche. Un primo lavoro ha analizzato la bioaccessibilità di vitamine, acidi fenolici e capacità antiossidante (TAC) in diverse frazioni di frumento duro ricche di strato aleuronico. I risultati hanno messo in luce che queste frazioni sono nutrizionalmente interessanti, grazie al loro contenuto in fibra e all’eccellente bioaccessibilità dei composti analizzati. In particolare, le vitamine, i composti fenolici e la capacità antiossidante sono risultati maggiormente bioaccessibili nella parte più interna dello strato aleuronico rispetto alla parte più esterna e alla crusca, rendendo la parte interna dell’aleurone interessante come ingrediente da utilizzare nella formulazione di nuovi prodotti. Un secondo tema del progetto ha riguardato lo studio dell’effetto della cottura (mediante bollitura e cottura a risotto) sulla capacità antiossidante e sul profilo fenolico di tre varietà di riso integrale differentemente pigmentate. La capacità antiossidante ha risentito maggiormente della bollitura in tutte le varietà, mentre solo nella varietà di riso nero la cottura a risotto ha determinato un incremento della TAC. I diversi composti fenolici hanno risentito in maniera diversa della cottura, con una diminuzione generale per antocianine, flavan-3-oli e flavonoli, mentre alcuni acidi fenolici non hanno risentito del trattamento termico. E’ stato inoltre valutato l’effetto della cottura sulla struttura anatomica del riso, osservando modifiche soprattutto nell’endosperma. E’ stato ipotizzato che durante la cottura parte dei composti, inclusi gli antiossidanti, vengano lisciviati nel liquido di cottura e che nel caso del risotto vengano successivamente riassorbiti, probabilmente attraverso le fratture della cariosside. Questo suggerisce che per una migliore comprensione dell’effetto della cottura sulla capacità antiossidante sia importante considerare non soltanto i composti antiossidanti ma anche la struttura della matrice.

The epidemiological evidences suggest that whole grain consumption is associated with a reduced risk of chronic diseases. This probably occurs because whole grains contain dietary fibre, micronutrients and antioxidant compounds, mostly located in the outermost layer of the caryopsis. However, the nutritional quality of a food does not depend only on the content of phytochemicals, but also on their bioaccessibility, which is influenced by the structure of the food matrix and the treatments (e.g. thermal, mechanical) done on food. From these considerations, the PhD project was developed in two topics. The first work analyzed the bioaccessibility of vitamins, phenolic acids and antioxidant capacity (TAC) in different aleurone wheat fractions. The results showed that these fractions are nutritionally interesting, thanks to their high fibre content and an excellent bioaccessibility of analyzed compounds. In particular, vitamins, phenolic compounds and the antioxidant capacity were more bioaccessible in the inner part of aleurone layer with respect to the outer part of aleurone and the bran. It suggests that the inner part of aleurone can be used as an ingredient in the formulation of new wheat based products. A second topic of the PhD project regarded the effect of cooking (with “risotto” method and boiling) on antioxidant capacity and phenolic profile of three differently pigmented rice varieties. The antioxidant capacity was affected by boiling in all the varieties, whereas risotto cooking caused an increase of the TAC only in black rice variety. Cooking differently influenced the different phenolic compounds, with a decrease of anthocyanins, flavan-3-ols and flavonols, whereas some phenolic acids were not affected. Moreover, the effect of cooking on the anatomical structure of rice was evaluated, observing modifications mainly in the endosperm. It was hypothesized that during cooking part of the compounds, included antioxidant compounds, are leached into the cooking medium and then, in the case of risotto cooking, they are subsequently re-absorbed, probably through the fractures in the caryopsis. This suggests that for a better comprehension of the cooking effect on antioxidant capacity it is important to consider not only antioxidant compounds, but also the structure of the matrix.
