dc.description.abstract | Derivati simmetrici dell'urea, quali N,N'-bis-(2,3-metilenediossifenil)urea (2,3-MDPU) e N,N'-bis-(3,4-metilenediossifenil)urea (3,4-MDPU), cioè MDPU, possono agire sia come coadiuvanti la radicazione avventizia che codiuvanti l'embriogenesi somatica, pur non avendo alcuna attività auxino- o citochinino-simile. In un tentativo volto a meglio comprendere le regule strutturali che sottendono l’azione adiuvante la radicazione esercitata dalle MDPU, sono stati sintetizzati altri derivati dell’urea nei quali un ossigeno del gruppo diossimetilenico delle MDPU è stato sostituito con un azoto per formare un gruppo benzossazolico. I nuovi derivati, la 1,3-di(benzo[d]ossazol-5-il)urea (5-BDPU) e la 1,3-di(benzo[d]ossazol-6-il)urea (6-BDPU), caratterizzati ancora una volta da una simmetria della molecola, in modo analogo alle MDPU non possiedono alcuna attività auxino- o citochinino-simile ma esercitano un effetto migliorativo della radicazione avventizia. Lo scopo di questo dottorato di ricerca è stato quello di studiare lo spettro d’azione delle BDPU e di meglio comprendere il loro meccanismo d’azione. A tal fine, l’attività adiuvante delle BDPU è stata studiata anche in altri processi fisiologici, nei quali sono coinvolti auxine e/o citochinine, quali l’embriogenesi somatica e la rigenerazione di germogli. dc.description.abstract | Symmetric urea derivatives, such as N,N’-bis-(2,3-methylenedioxyphenyl)urea (2,3-MDPU) and the N,N’-bis-(3,4-methylenedioxyphenyl)urea (3,4-MDPU), namely MDPUs, can act either as adventitious rooting adjuvants or as somatic embryogenesis adjuvants. These compounds do not show any auxin- or cytokinin-like activity per se, rather it seems that they magnify the response to the growth regulator stimulus of naturally competent-to-root or -to-form-somatic-embryos cell niches. When a chemical modification of the aromatic moiety of MDPUs consisting in a replacement of the methylenedioxyphenyl groups by the isostere benzoxazole ones was performed, the two new compounds, the 1,3-di(benzo[d]oxazol-5-yl)urea (5-BDPU) and the 1,3-di(benzo[d]oxazol-6-yl)urea (6-BDPU), namely BDPUs, showed a conserved adventitious rooting enhancement without either auxin- or cytokinin-like activity per se, as already demonstrated by the lead compounds. It has been hypothesized the existence of a new category of adventitious rooting adjuvants, i.e. symmetrical urea derivatives showing similar chemical structure, interacting with endogenous or exogenous auxin, without being auxin synergists. Here we report new insights about the adjuvant activity of BDPUs in several physiological processes, in which auxins and/or cytokinins are involved, as adventitious rooting, somatic embryogenesis and shoot regeneration. In addition, here we describe our findings regarding the interaction among BDPUs and the cytokinin signalling pathway. | it |
