dc.description.abstract | La traslocazione t(9;11)(p22;q23) genera l’oncogene MLL-AF9 ed è comunemente associata con la leucemia acuta mieloide monocitica (LAM-M5; classificazione FAB). Per l’oncogenicità di MLL-AF9, è richiesta l’(over)espressione di diversi altri geni, tra cui HOXA4, 5, 6, 7, 9, 10 e 11 (definiti come “HOXA-code” genes), MEIS1 (un cofattore delle proteine “HOXA-code”) e MYB. Precedentemente, abbiamo riportato che la down-regolazione dell’espressione di MLL-AF9 non era obbligatoria per la maturazione monocito-macrofagica in cellule di LAM-M5 con traslocazione t(9;11)(p22;q23). In questo studio abbiamo analizzato l’espressione dei geni “HOXA-code”, MEIS1 e MYB, mediante PCR semi-quantitativa Real Time, durante il differenziamento monocito-macrofagico indotto dal phorbol 12-myristate 13-acetate (PMA), in cellule THP-1 che possiedono la traslocazione t(9;11)(p22;q23) ed esprimono MLL-AF9. I risultati mostravano che tutti i geni analizzati erano espressi nelle cellule THP-1 non trattate. Dopo l’induzione del differenziamento, abbiamo osservato una down-regolazione di HOXA4, 7, 10, 11, MEIS1 e MYB, indicando che l’espressione di questi geni potrebbe essere implicata nel blocco del differenziamento osservato in leucemie relative a MLL-AF9. Poiché la chemioterapia standard non è curativa per molti pazienti con LAM-M5 con anormalità 11q23/MLL, abbiamo anche valutato gli effetti di diverse molecole, singole ed in combinazione, a concentrazioni clinicamente ottenibili, sul differenziamento lungo il lineage monocito-macrofagico, in linee cellulari di LAM-M5 con traslocazioni t(9;11)(p22;q23) e che esprimono MLL-AF9. I risultati indicavano che la combinazione di agenti demetilanti, come la decitabina, e molecole che inducono il differenziamento monocitico normale, come la Vitamina D3, aveva una forte attività anti-leucemica e si raccomandano ulteriori investigazioni di tale combinazione in questo tipo di neoplasia. | it |
dc.description.abstract | The translocation t(9;11)(p22;q23) generates the MLL-AF9 oncogene and is commonly associated with monocytic acute myeloid leukemia (AML-M5; FAB-classification). For the oncogenicity of MLL-AF9, the (over)expression of several other genes, including HOXA4, 5, 6, 7, 9, 10 and 11 (defined as “HOXA-code” genes), MEIS1 (a cofactor of “HOXA-code” proteins) and MYB is required. We previously showed that the down-regulation of MLL-AF9 expression is not obligatory for monocyte-macrophage maturation in AML-M5 cells carrying t(9;11)(p22;q23). In this study, we analyzed the expression patterns of “HOXA-code”, MEIS1 and MYB genes, by semiquantitative Real Time PCR means, during the monocyte-macrophage differentiation induced by phorbol 12-myristate 13-acetate (PMA), in THP-1 cells carrying t(9;11)(p22;q23) and expressing MLL-AF9. The results showed that all the analyzed genes were expressed in untreated THP-1 cells. After the induction of differentiation, we observed a down-regulation of HOXA4, 7, 10, 11, MEIS1 and MYB, indicating that the expression of these genes could be implicated in the differentiation blockage observed in leukemias with 11q23/MLL abnormalities. Because standard chemotherapy is not curative for many patients with AML MLL-related, we also evaluated the effects of several molecules, alone or in combination, at clinically achievable concentrations, on differentiation along monocyte-macrophage lineage, in AML-M5 cell lines carrying t(9;11)(p22;q23) and expressing MLL-AF9. The results indicated that strategies combining demethylating agents, such as decitabine, and drugs that induce normal monocytic differentiation, such as Vitamin D3, had strong anti-leukemic activity and warrants further investigation in this type of malignancy. | it |
